
Samyang Biopharm announced on the 20th that nonclinical study results for its proprietary gene delivery platform, "NanoReady," designed for therapeutic mRNA cancer vaccines, have been published as the cover article in the Journal of Controlled Release.
NanoReady is a core technology within Samyang Biopharm’s SENS (Selectivity Enabling Nano Shell) platform, combining in-house developed lipids with biodegradable polymers to create advanced nanoparticle delivery vehicles. The system is engineered to selectively deliver mRNA to dendritic cells in the spleen, which play a central role in immune function by presenting cancer antigens to T cells, prompting them to attack tumor cells.
A primary advantage of NanoReady is its "ready-made" design. The delivery system can be manufactured and stored in advance without mRNA. Once patient-specific cancer antigen mRNA is synthesized based on individual tumor characteristics, a personalized vaccine can be assembled rapidly by simply mixing the mRNA into the carrier.
Unlike conventional lipid nanoparticles (LNPs), NanoReady features a unique multilayer structure capable of loading approximately 20 times more mRNA per particle. This significantly higher payload induces a powerful T-cell immune response against targeted cancer cells.
Furthermore, traditional LNP formulations often rely on polyethylene glycol (PEG)-lipids and phosphatidylcholine (PC)-based helper lipids, which can trigger hypersensitivity reactions or rapid drug clearance upon repeat administration. NanoReady eliminates PEG-lipids and PC-based helper lipids entirely, optimizing its composition to support safe, repeated dosing—a crucial requirement for cancer therapies that require multiple cycles.
In animal studies, NanoReady loaded with tumor antigen mRNA selectively targeted splenic cells and generated strong antigen-specific T-cell responses. Comprehensive blood and tissue evaluations following repeated administration revealed no signs of systemic toxicity, providing solid nonclinical evidence for the safety of multi-dose regimens.
Samyang Biopharm noted that the results demonstrate how a single delivery platform can fulfill two critical prerequisites for commercializing personalized mRNA cancer vaccines: rapid, scalable manufacturing and high immunological efficacy.
"This achievement is significant because it provides solid preclinical evidence that NanoReady is not merely a theoretical concept, but a versatile platform adaptable to a wide range of therapeutic mRNA candidates," a Samyang Biopharm official said.
The study was published under the title "NanoReady: A ready-made DC-selective delivery platform for personalized mRNA cancer vaccines with strong NHP immunogenicity."
